What is the difinative moment of humor in the OT?
I'm gonna have to go with a scene in ANH....
Han Solo decides to impress Luke and Leia by charging down a hall in the Deathstar, a valiant battle cry upon his lips, when, seconds later, we hear the same battle cry, pitch, octive, decibel level, all the same, however this time it is a cry of "blah! help! im gonna sting in the morning!" because he charged his way right into a patrol of storm troopers! ;D Gets me to laugh every time even though I know its gonna happen.

One of my favorite "comical" scenes is in ROTJ while Han Solo and Luke Skywalker are being taken to the sarlaac pit......
Han: "I think my vision is getting better.....instead of seeing a big dark blur, I'm seeing a big light blur."
Like: "I used to live here, ya know."
Han: "You're gonna die here, ya know."
My second is when Luke struggles with Yoda (unknowingly at the time) to get his light back and Yoda says -
"Mine! Or I will help you not!"
Then he keeps smacking R2 trying to get him to let go of the light screaming -
"Mine! Mine! Mine!"

For me it was most of the comic relief provided by C-3PO- my two favourites are both in ANH:
After R2 shuts down the trash compactor and our heroes are shouting with joy, 3PO's line "Listen to them, they're dying R2!"
After the TIE fighter attack on the Millennium Falcon, with 3PO on the floor covered in wires yelling, "Help! I think I'm melting! This is all your fault!"

Unlike in the prequels, Threepio is generally very funny in the OT.
My favourite funny bits are:
"And I thought they smelt bad... on the outside!"
Just now I'm trying to think of funny bits... and I've realised that what makes the parts funny is the way that the actors deliver the lines. Especially Harrison Ford and Anthony Daniels, they were fantastic in the OT.

I'm gonna have to disagree with you on one item of C-3PO's humor in the PT. I think that while the puns were obvious and predictable, that the TIMING of them are perfect in the Geonosis battle scene

There isn't any single moment of definitive humor in OT. It's sprinkled around in various places. I like the scene where 3PO tells Han that it's against his programming to impresonate an diety in ROTJ. Look at lLuke during that exchange; very funny. R2D2 and C3PO were main vehicles of humor in the OT, but I'm glad they were less so in the prequels; one can over-do a joke.

When Leia tells Luke to find the controls that extend the bridge and Luke admits that he just blew them up....that never seems to get a laugh, but I think it's funny. Maybe it's the timing.
In the prequels....the bit with Qui-Gon and Jar-Jar's tongue; that's funny. And love it when Annakin's lightsaber gets destroyed in the droid factory and he just looks depressed and says, "No again. Obi-Wan's gonna kill me." And when Yoda confronts Darth Sidious and just brushes the guards aside on his way in....that always gets a laugh.

I would have to say that Yoda taking out the Royal Guards is the funniest moment of the entire saga for me!

one of my favorites is Solo in ROTJ when he is about to sneak up on the biker scouts:
"Hey, it's me!" said with a thingyy grin...

OT: Almsot all of Han's smarmy remarks. Han's reaction when he realises Leia had a brother. "He's your brother?!" LOL hehehe.
PT: Qui and jar jar. R2D2's antic. Epsically in ROTS. Anikan and Obi Wan cracking about R2.
R2 and Obi Wan chatting via comlink, he sees droids approaching and stuffs the 'link into his innards. of Obi wan coming from within saying. "R2. Can you hear me? We need to go up not down. R2?" The poor astromech must've been a bit exasperiated at that point.
ANother incident: Docking bay, R2's dealing with a couple of battle droids. he squirts oil on the floor and inginights it. BBQue anyone? :-D

Han had the best lines. "I know" (responding to Leia confession that she loves him) is by far the one line that defines his character, and was delivered perfectly. ;D
Han's exchange over the intercom (just after he, Luke and Chewie have shot up the prison control room aboard the Death Star) is another great moment in humour.
"Uh... everything's perfectly all right now. We're fine. We're all fine here now, thank you. How are you?"
That and...
"Better her than me!"
Oh here's another moment...
Han shoots Greedo, who falls face first on the bar table- Everyone in the bar turns to look and Muftak scratches the side of his furry head as if to say, "Huh? What happened?"
